射麻止喘液对哮喘大鼠气道平滑肌细胞作用研究

摘　　要：目的:探讨哮喘大鼠模型建立及其气道平滑肌细胞培养方法,了解其生长特性,通过研究射麻止喘液中药血清对气道平滑肌细胞增殖效应,为研究哮喘病预防及治疗提供理论及实验支持。方法:制作慢性哮喘大鼠模型,用酶联法培养哮喘大鼠气道平滑肌细胞,倒置相差显微镜观察其生长情况,MTT检测生长曲线。结果:以酶联法培养7天可见细胞贴壁,14天左右融合90%左右。传代后细胞"谷峰"生长明显。结论:本实验提供了一种常见简单的哮喘模型,培养的细胞为平滑肌细胞,射麻止喘液血清具有抑制哮喘大鼠气道平滑肌细胞增殖作用,低剂量药物血清组相对于中高剂量组抑制作用更强。Objective：To probe into the establishion of asthmatic rat model and the method of the cells,cultivation of asthmatic airway smooth muscle,to know theirs growth characters,through researching the poliferative effection reply into Shemazhichuan liquid drug serum,we provide theoretical and experimental support for studying the prevention and treatment of asthma.Method：To production of chronic asthmatic rat model,through ELISA method cultiveing asthmatic rat airway smooth muscle cells,observing the growth of the cells via MTT and detecting theirs growth curve.Result：Cell Adherent after seven days cultivation via ELISA method,about 14 days probably 90% of cells fusion,the growth of after passage cells significantly was ＂valley-Peak＂.Conclusion：This experiment provides a common simple model of asthma rat,cultured cells as smooth muscle cells,Shemazhichuan liquid indeed can inhibit the asthmatic airway smooth muscle cells,and the proliferation of serum low-dose group relative to high dose group has a stronger inhibition.
