伊贝沙坦对人肾系膜细胞β1-整合素表达及Ⅳ型胶原分泌的影响

摘　　要：目的探讨伊贝沙坦对糖基化终产物(AGEs)干预下人肾系膜细胞(HRMC)β1-整合素表达以及胞外基质(ECM)成分Ⅳ型胶原(Ⅳ-C)分泌的影响。方法用糖基化终产物-牛血清白蛋白(AGE-BSA)干预HRMC(AGE-BSA组),用不同浓度的伊贝沙坦与AGE-BSA共同干预HRMC48 h(伊贝沙坦干预组)。RT-PCR法测定细胞β1-整合素mRNA水平;Western blot法检测β1-整合素蛋白表达;放射免疫分析法测定细胞培养上清Ⅳ-C含量。结果与AGE-BSA组相比,伊贝沙坦干预组能下调由AGE-BSA诱导的HRMCβ1-整合素mRNA和蛋白表达的升高,并抑制培养上清Ⅳ-C的分泌(P<0.05)。结论 伊贝沙坦可能通过抑制AGEs引起的上述改变而发挥其肾脏保护作用。Objective To investigate the effects of irbesartan on the expression of β1-integrin and the secretion of type Ⅳ collagen（Ⅳ-C） of cultured human renal mesangial cells（HRMCs） incubated with AGEs.Methods HRMCs were incubated with different concentrations of irbesartan with AGE-BSA for 48 hours.The mRNA expression of β1-integrin in HRMC was analyzed by RT-PCR,the protein expression of β1-integrin in HRMCs was determined by Western blot.The content of Ⅳ-C in cell culture media was measured by radioimmunoassay.Results Irbesartan decreased the levels of β1-integrin mRNA and protein in HRMCs and the level of Ⅳ-C in supernatants of cultured HRMCs incubated with AGE-BSA（P0.05）.Conclusion Irbesartan has renal protection,which may be via supression of AGEs.
