泽泻汤对代谢综合征大鼠神经肽及其Y1受体的影响

摘　　要：目的观察泽泻汤对代谢综合征(MS)大鼠血浆神经肽(NPY),下丘脑NPY及其Y1R表达的影响,探讨其可能机制。方法用高糖高脂饲料喂饲建立MS大鼠。将23只MS大鼠分为3组,分别给盐水、泽泻汤、盐酸西布曲明。4 w后,测体重、血糖、三酰甘油(TG),放免法检测血浆NPY;免疫组化法检测下丘脑NPY及其Y1R表达。结果与盐水组相比,泽泻汤组大鼠体重、血糖、TG、血浆NPY水平、下丘脑NPY及其Y1R表达的平均光密度均显著降低(P<0.05)。结论泽泻汤对MS大鼠有良好的治疗作用,其机制可能与抑制下丘脑NPY及其Y1R表达有关。Objective To observe the effect of Zexie Tang on plasma neuropeptide Y（NPY） and hypothalamus NPY Y1 receptor in metabolic syndrome（MS） rats and the possible mechanism.Methods MS rats were made by feeding high sugar fat diet.23 MS rats were randomly divided into salt water,Zexie Tang and Sibutramine groups.Body weight,glucose（Glu）,triacylglyceride（TG）,and plasma NPY of the rats were measured after four weeks.Meanwhile,NPY Y1R in hypothalamus were tested by immunohistochemistry.The density mean was analyzed by image-pro plus 6.0.Results Zexie Tang could inhibit the growth of body weight and reduce the levels of Glu,TG and plasma NPY（P0.05）.NPY Y1R in hypothalamus were significantly lower in Zexie Tang group than those in the saltwater group（P0.05）.Conclusions There is obvious therapeutic effect of Zexie Tang on MS rats,which is possibly related to its inhibition of the synthesis of NPY and the expression of Y1R in hypothalamus.
