摘 要:[目的]观察降钙素对去卵巢大鼠腰椎骨量和椎间盘退变的作用。[方法]30只3个月龄Sprague-Dawley(SD)大鼠,随机均分为3组,每组10只:假手术(Sham)组,去卵巢(OVX)组,去卵巢+降钙素(OVX+CT)组。3个月后处死动物,并于处死前10d和4d分别给予四环素和钙黄绿素双荧光标记。取L2~4椎体进行骨密度及骨组织形态计量学分析,L4~5腰椎及椎间盘进行VG染色,运用免疫组化检测椎间盘MMP-13的表达,观察椎间盘的病理学改变并根据组织学评分系统对腰椎间盘的退变程度(LVD)进行评分。[结果](1)腰椎间盘评分:OVX+V组显著高于Sham组及OVX+CT组;(2)腰椎骨密度及骨形态计量学骨量参数(BV/TV,Tb.N):OVX+V组显著低于Sham组及OVX+CT组;(3)免疫组化:OVX+V组大鼠椎间盘中基质金属蛋白酶MMP-13表达显著高于OVX+CT组、Sham组。[结论]降钙素可预防去卵巢大鼠腰椎间盘的退变,其作用机制可能与维持椎体骨量及微观结构,同时降低椎间盘中MMP-13的表达抑制基质的降解有关。[Objective]To investigate the effects of calcitonin on bone mass and lumbar intervertebral disc degeneration(LVD) in the ovariectomized(OVX) rats.[Method]Thirty 3-month-old female Sprague-Dawley rats were randomized into three groups.Ovariectomy was performed in 20 rats and the other group of 10 rats was subjected to Sham surgery(Sham group).One group of OVX rats was injected subcutaneously with salmon calcitonin(20 IU/kg/d)for 3 months(CT group).Another group of OVX rats(OVX group) and the Sham group were treated with vehicle.Double labeling was performed with subcutaneous injections of tetracylcin 10 days and of calcein 4 days before sacrifice.Bone mineral density(BMD) measurement and bone histomorphometric analysis were applied to the segments of L2~4 lumbar spines in all rats killed 3 months of postsurgery.L4-L5 lumbar spines and intervertebral disc sections were stained with collagen VG staining,and immunostained with MMP-13.The pathological changes of intervertebral discs were observed and the LVD degree was scored by a histological scoring system.[Result](1)The histological score for LVD in OVX group was markedly increased compared with those in Sham and OVX+CT groups.(2) The BMD and histomorphometric parameters of bone mass(BV/TV) of rats in both OVX+CT and Sham groups were significantly higher than that of OVX group.(3) The expression of MMP-13 in the OVX group was significantly higher than those in the Sham group and OVX+CT group.[Conclusion]CT could suppress lumbar intervertebral disc degeneration in the OVX rats.The mechanism may be associated with the prevention of bone loss and deterioration of microarchitecture,and the inhibiting of the degraded matrix by downregulation of MMP-13.
