壳聚糖对大鼠Ⅲ度烧伤创面愈合及脂质过氧化反应的影响

摘  要:目的:研究壳聚糖对大鼠Ⅲ度烧伤创面脂质过氧化反应的影响及在创面愈合中的意义。方法:按Nagelschmidt法计算大鼠Ⅲ度烧伤后各时相点创面愈合率;用生化法测定各时相点创面组织蛋白、羟脯氨酸(HOP)、超氧化歧化酶(SOD)、丙二醛(MDA)含量。结果:4%壳聚糖组(w/v)各个时间点创面愈合率都明显高于自然愈合组(P<0.01);伤后7,14d,4%壳聚糖组织蛋白含量明显高于自然愈合组(P<0.01或P<0.05);2%壳聚糖组、4%壳聚糖组烧伤后7d HOP就达最高值,且其值比自然愈合组最高值大;伤后3,7,14d,2%壳聚糖组、4%壳聚糖组创面SOD活性比自然愈合组高(P<0.05);4%壳聚糖组创面MDA含量的升高比自然愈合组明显低(P<0.01或P<0.05)。结论:壳聚糖对大鼠Ⅲ度烧伤创面有促愈合作用,并呈剂量依赖性,其促愈合机制与使用壳聚糖后创面脂质过氧化反应程度减轻有关。OBJECTIVE To study the effects of chitosan on lipid peroxidation in the wound of Ⅲ degree skin burns in rats. METHODS The wound healing rate was investigated on the burned wound of rat by way of Nagelschmidt's. The contents of protein, hydroxyproline(HOP), malondialdehyde (MDA), and the activity of superoxide dismutase (SOD) were measured. RESULTS The wound healing rate of 4% chitosan group(w/v) was higher than that of the normal group. At the 7th, 14th day postinjury, the content of protein of 4% chitosan (w/v) group was higher than that of the normal group. The content of HOP of 2 % and 4% chitosan (w/v) group was highest at the 7th day postinjury. At the 3rd, 7th and 14th day postinjury, the decressing degree of the activity of SOD and the increasing degree of MDA in 4% chitosan group(w/v) were less than those of the other groups(P〈0. 01 and P〈0. 05 ) . CONCLUSION Chitosan could promote the wound healing of Ⅲ degree skin burns in rats significantly, in a dose-depentent manner. Its mechanism was probably ralated to chitosan relieving lipid peroxidation of the burn wound.
