摘　　要：目的:研究雷至胶囊对阿霉素肾病大鼠肾脏nephrin蛋白表达的影响,探讨其抗蛋白尿的作用机制。方法:采用分次尾静脉注射阿霉素建立阿霉素肾病模型,将实验动物分为模型组、雷公藤多苷组、雷至胶囊组及正常对照组,第5周末处死大鼠,光镜、电镜观察肾组织病理形态学变化;半定量RT-PCR和Western-blot的方法检测肾皮质中nephrin mRNA和蛋白的表达。结果:与模型组相比,治疗组24h尿蛋白、血脂明显降低,同时nephrin mRNA及蛋白表达明显提高,透射电镜显示足细胞足突病变不同程度改善;雷至胶囊组以上指标改善明显优于雷公藤多苷组,且无肝脏副作用。结论:nephrin蛋白的表达减少是产生蛋白尿的主要原因;雷至胶囊抗蛋白尿的作用机制之一是提高肾小球细胞nephrin蛋白的表达,从多方面证实其治疗效果优于雷公藤多苷。OBJECTIVE To investigate the effect of Leizhi capsule on nephrin mRNA in rats with adriamycin（ADR） in duced-nephropathy and to explore the mechanism of Leizhi capsule on proteinuria. METHODS ADR nephropathy was induced by tail intravenous injection of adriamycin in day 1 and day 7. The rats were divided into model group, the multiglycosides of Tripterygium wilfordii Hook（GTW） group, Leizhi capsule group and the normal group. At the end of the 5th week, the rats were sacrificed and the renal tissue was acquired for the examinations of light microscopy, electron microscopy. The mRNA and protein expression of nephrin were examined by RT-PCR and Western-blot respectively. RESULTS ADR nephropathy was induced by tail intravenous injection of adriamycin in day 1 and day 7. The rats were divided into model group, the multiglycosides of Tripterygium wilfordii Hook（GTW） group, Leizhi capsule group and the normal group. CONCLUSION （1）The decreased expression of nephrin was one of molecular mechanism that leads to the occurrence and development of proteinuria in ADR rats. The increasing nephrin expression may be one of the mechanisms of anti proteinuria of GTW; （2）The effect of Leizhi capsule was superior to GTW in multi-respects.
