摘 要:通过基因组步行技术克隆到长春花脱乙酰氧基文多灵-4-羟化酶基因上游905bp启动子序列,生物信息学分析发现该序列中存在多个与光照诱导有关的顺式元件,同时也发现一个与长春花特异的顺式作用元件JERE类似的短序列,推测该序列是茉莉酸诱导途径中转录因子ORCA3的特异结合位点。Desacetoxyvindoline-4-hydroxylase(D4H) is an important enzyme in the biosynthetic pathway of monoterpenoid alkaloids in Catharanthus roseus.It catalyzes the 4-hydroxylation of desacetoxyvindoline to deacetylvindoline.The aim of this work was to clone and to investigate the functional profiles of d4h promoter.The C.roseus d4h promoter sequence was isolated by a PCR-based genome walking method.As a result,a 905bp promotor sequence was cloned based on the cDNA sequence of d4h from Genbank.Sequence analysis using PlantCARE online database showed several cis-acting elements,including several light-induction-involving elements.It demonstrates the mechanism of the light to vindoline biosynthesis.In addition,a short region similar to the C.roseus cis-element JERE was also found.We speculate that this region may be the binding site of the C.rosues transcription factor ORCA3.
