摘  要:通过电化学氧化利血平,使其生成波长为495 nm的强荧光物质3,4-二去氢利血平,实现了药物中利血平含量的荧光检测.研究结果表明,电化学氧化利血平后产物的荧光强度与样品中的利血平浓度分别在6.00×10-8~2.00×10-6mol/L和2.00×10-6~8.00×10-5mol/L范围内呈良好的线性关系,检测限为3.90×10-8mol/L.该方法用于药物制剂中利血平含量的测定,相对标准偏差小于2.33%(n=5),加标回收率为96.6%~104%.A simple and sensitive fluorescence method has been developed by combining electrolysis and fluoremetry for the determination of reserpine.This method is based on that reserpine was oxidized at a platinum electrode,and its product has a characteristic fluorescence at 495 nm with excitation wavelength at 385 nm.The electrochemical reaction conditions and fluorescent experiment parameters were studied and optimized.Under the optimum conditions,the fluorescent intensity of the electrolyzed product was proportional to the concentration of reserpine over the range from 6.00×10-8 to 2.00×10-6 and 2.00×10-6 to 8.00×10-5 mol/L,with the correlation coefficients of 0.999 0 and 0.999 5,respectively.The detection limit was estimated to 3.90×10-8 mol/L.The relative standard deviation was less than 2.33%(n=5) and the recovery was in the range of 96.6%~104% for the determination of reserpine in pharmaceutical samples.The proposed method showed a rapid and sensitive advantage to detect reserpine.Especially,the application of electrochemical oxidation could effectively avoid the effect of reducing agent,providing a green technique for the determination of reserpine.
