日粮诱发豚鼠胆囊炎的病理学及酶组化研究

摘  要:本研究采用高脂肪日粮;高胆固醇日粮;高碳水化合物、低脂、低蛋白日粮;低蛋白、低热卡日粮以豚鼠为动物模型诱发其胆结石形成,通过酶病理组织化学技术结合胆汁成份分析,探讨了多种酶在胆结石形成过程中的作用。结果表明:4 种致石日粮都能造成豚鼠胆囊炎症和结石形成,肝脏表现为胆管炎和动脉炎,胆囊及肝外胆管粘液腺显著增生。胆囊粘膜β G C D、 A C P、 A K P 活性升高, G6 Pase 活性下降,且各实验组间无明显差异。肝脏β G C D 变化不明显, A C P、 A K P、 G6 Pase3种酶活性变化明显,且各实验组间差异较大。胆汁理化成份明显改变:胆汁β G C D 活性升高且与胆囊炎的严重程度有关,胆汁中 T B、 C B、 U C B、 Ca2+ 的含量均升高,并且胆汁各理化成份之间有一定的相关关系。通过分析认为:胆结石的形成与酶代谢紊乱,尤其是β G C D 的紊乱和胆汁中各成份含量失衡有密切关系。Using enzymic histopathology and histochemistry techniques assited with bile analysis we studied the effect of enzyme metabolism disorder and mucopolysaccharids in the gallstone formation which induced by diets of high fat;high cholesterol;high carbohydrate,low fat and low protein;low protein and low calory on guinea pigs. The result showed that:four different diets could cause cholecystitis and gallstone,β GCD activity in the mucosa of gallbladder increased obviously but slightly in the livers.ACP,AKP and G 6-Pase activity changed clearly in the gallbladder and liver.β GCD activity in bile had relation to cholecystitis.TB、CB、UCB、Ca 2+ concentration in bile increased and these ingredients had some regression relitionship.We concluded that gallstone formation had a close relationship to enzyme metabolism disorder,especially β GCD activity.
