摘  要:目的探讨CYP2E1基因多态性对苯乙烯代谢的影响。方法以56名苯乙烯接触工人为研究对象,应用个体采样器测定其个体接触剂量,应用高效液相色谱法测定其尿中主要代谢产物(苯乙醇酸和苯乙醛酸)的含量,应用聚合酶链反应-限制性片段长度多态性分析技术(PCR-RFLP)检测其CYP2E1基因5端上游调节区R saI位点及第六内含子区D raI位点的多态性。结果CYP2E1基因5端R saI多态性对职业接触苯乙烯低浓度组工人尿中苯乙醇酸和苯乙醛酸含量均有显著影响(P<0.05)。结论CYP2E1基因5端R saI多态性对苯乙烯代谢结果具有一定影响,可能是苯乙烯职业接触遗传易感性因素。Objective To explore the influence of genetic polymorphisms of CYP2E1 on the metabolism of styrene.Methods 56 styrene-exposed workers were selected as study subjects.Personal passive samplers were used for determining personal time-weighted average(TWA),HPLC was used for detecting its main metabolites (mandelic acid and phenylglyoxylic acid) in urine,and polymerase chain reaction-restrained fragment length polymorphism technique(PCR-RFLP)was applied to detect the single nucleotide polymorphisms (SNPs)on CYP2E1(5-flanking region,RsaI,and intron 6,DraI).Results There was a significant association between urinary metabolites and genotypes of CYP2E1(5-flanking region,RsaI)in low-exposed group(P0.05).Conclusion Styrene metabolism could be influenced by genotypes of CYP2E1(5-flanking region,RsaI),this kind of genotypes can be regarded as susceptible factor,that might be helpful in evaluating the individual susceptibility and using in screen of styrene-exposed populations.
