摘  要:采用改进的水稻病毒纯化方法,以200 g/L的蔗糖垫替代蔗糖密度梯度,从RStV 感染的水稻叶片中制备了病毒颗粒.电镜观察显示,RStV 为细丝状和分枝状结构.变性琼脂糖凝胶电泳分析分离纯化的病毒总RNA得到4 个大小不同的病毒RNA.SDSPAGE可检测到病毒编码的外壳蛋白.提纯病毒作为抗原与所制备的抗血清及日本抗血清均有血清学反应.Rice stripe virus (RStV) from infected rice leaf was purified by a modified procedure of conventional purification with 200 g/L sucrose cushion instead of linear sucrose gradient. The virus with filamentous and branched filamentous particles were observed in purified preparations under electron microscope. Significant size differences were found in the single stranded forms of genomic RNAs of RStV by denatured agarose gel electrophoretic analysis. The viral coat protein was detected in SDS PAGE. With the antibody of RStV form Japan,the virus showed a serological homology. Thus,it indicates that this simple protocol is suitable for purifying the RNP particles of RStV.
