摘　　要：目的:观察胎儿头皮毛囊中CD34的表达。方法:获取因胎儿畸形引产的胎龄8个月的死亡2 h胎儿头皮,分成2份。1份制备冷冻切片,CD34免疫荧光标记,激光共聚焦显微镜观察摄片;另1份无菌处理,消化头皮获取游离毛囊,制备毛囊细胞悬液,采用流式细胞术检测。结果:共聚焦显微镜下观察到,CD34主要表达于胎儿头皮的毛囊外根鞘隆突区,也见于毛球处、表皮基底部;流式细胞仪检测出CD34+细胞占毛囊总数的8.54%～13.04%。结论:胎儿头皮毛囊隆突区有较多的CD34+细胞,推测该部位为毛囊干细胞的居处。部分毛囊中部、毛球处出现CD34+细胞,提示不同毛发周期,干细胞或其直接子代的居所发生变化。胎儿表皮基底层存在均匀分散的CD34+细胞。Objective：To observe the expression of CD34 in fetal scalp.Methods：The fetal scalp with follicles was from a fetus（at eight months of gestational age） within 2 h of his death after an abortion due to abnormal development.The tissue was divided into two copies.An equal part was prepared for the frozen sections labeled by CD34 immunofluorescence and observed by laser confocal microscope.Another part was digested by collagenase type Ⅱ for harvest of free hair follicles,which were furtherly treated by trypsin for cells suspension.The hair follicle cells were assayed by flow cytometry.Results：CD34 was mainly expressed in the outer root sheath bugle area of hair follicle under confocal microscopy,a minority was seen in the hair bulb and the basilar part of epidermis.The data from flow cytometry demonstrated that CD34 positive cells accounted for 8.54-13.04 percentages of total isolated follicle cells.Conclusions：Most CD34 positive cells are localized at the bulges of hair follicle which is presumed as the residence of follicle stem cells.The presence of CD34 positive cells in the follicular middle part and lower part around hair bulb,suggests the locations of stem cells and their immediately progenies varied along with the hair cycle.Furthermore,CD34 positive cells equably scatter at the basilar part of epidermis in the fatal scalp.
