摘 要:利用玻璃化保存方法,在自行设计的细胞渗透性实验台上,实时观察了黏附成骨细胞在高浓度玻璃化保护液中的形态变化.经过七步添加和七步去除95%的VEG玻璃化保护液后,黏附的成骨细胞的形态由细长的纺锤形变为粗短的圆柱形,说明黏附细胞的两端及边缘的黏附点已脱落,但经过3~4 h的培养后,黏附细胞逐渐恢复了原来的形态,表明玻璃化保护液的处理只是改变了细胞的形态,没有造成黏附细胞的脱落和严重损伤.The morphology changes of attached osteoblast cells exposed to vitrification solutions of various concentrations were observed by use of a diffusion stage.After seven steps of sequential addition and removal of 95% VEG,the morphology of attaches cells changed from a long podium or spindle-like appearance to short cylinder shape.The cells appear to lose contact with the scaffold at the periphery of the cell,releasing focal adhesions and becoming more centralized.When incubated for 3~4 h,the attached cells recover and retain their original morphology.The results indicate that vitrification solution treatment changes only the morphology of attached cells,but does not damage the cells or cause detachment of cells.The results of the paper provide information for the cryopreservation of attached cells by vitrification.
