摘 要:目的研究新型可注射温度敏感型聚乙二醇-聚乳酸-聚乙二醇水凝胶[poly(ethylene glycol)-poly(epsilon-caprolactone)-poly(ethylene glycol),(PEG-PCL-PEG,PECE)]的生物相容性,探讨其做为细胞三维培养材料和组织工程支架的可行性。方法将兔骨髓基质干细胞(bone marrow mesenchymal stem cells,BMSCs)接种于水凝胶,体外共同培养后进行形态结构观察,用MTT法检测BMSCs增殖活性,DAPI和EI试剂盒观察细胞存活和凋亡情况。结果 BMSCs在PECE水凝胶中生长及功能均比较理想。DAPI/EI染色见少量红色荧光,显示仅有少量细胞凋亡。结论可注射温度敏感性PEG-PCL-PEG水凝胶生物相容性好,是一种良好的三维培养材料,可作为种子细胞的载体应用于组织工程中。Objective To study the biocompatibility of a novel thermosensitive injectable[poly (ethylene glycol) -poly(epsilon-caprolactone) -poly(ethylene glycol),PEG-PCL-PEG(PECE)] hydrogel and investigate the possibility of PECE hydrogel as scaffold materials in tissue engineering. Methods BMSCs were cultured in PECE hydrogels in vitro,cell morphology was investigated and function cell proliferation was observed with MTT method,cell apoptosis were measured by using DAPI/EI kit.Results BMSCS cells were attached and extended well on hydrogel,cell viability were not affected.DAPI/EI double-staining showed little red fluorescence,which implied small amount of apoptosis when cells were cultured in hydrogel.Conclusion The injectable thermosensitive hydrogel is of good biocompatibility,and can be used as a scaffold material and the carriers of seed cells in tissue engineering.
