摘  要:目的探讨表皮葡萄球菌(SEP)耐药性与生物膜之间、PIA及ica操纵子各基因表达的关系。方法采用K-B法检测表皮葡萄球菌对抗菌药物的耐药性检验:刚果红琼脂平皿检测PIA,微量半定量法检测生物膜表型,PCR法和RT-PCR方法扩增ica操纵子,统计分析ica操纵子与PIA及生物膜之间的相关性。结果 49株SEP中对苯唑西林耐药34株(69.4%),敏感15株(30.6%),PIA与耐药密切相关差异有统计学意义(P<0.05),生物膜检测结果,32.7%有生物膜形成,67.3%无生物膜形成;ica A与PIA有密切相关性差异有统计学意义(P<0.01),ica A阳性10株,ica D阳性11株,ica A与ica D差异无统计学意义,ica R阳性7株,分别与ica A与ica D比较,无密切相关性,差异无统计学意义。结论表皮葡萄球菌对苯唑西林产生耐药性,耐药性与生物膜及PIA的形成有关,icaA和icaD基因的联合表达是PIA合成的关键环节,icaR基因的表达抑制PIA的合成。OBJECTIVE To explore the relationships between drug resistance of Staphylococcus epidermidis and biofilm formation,the positive rate of PIA production and the genic expression of Ica operon.METHODS The antibiotics susceptibility was tested by K-B method.Congo red agar plate was used to test PIA synthesis.Semi-quantitative test was performed to examine biofilm phenotype.IcaA gene and icaD were detected by polymerase chain reaction(PCR) and RT-PCR.RESULTS Among 49 strains,10 strains were sensitive to Oxacillin,39 strains were resistant to Oxacillin.There was relation between PIA and antibiotics susceptibility.(P0.05).32.7% of S.epidermidis strains were slime forming.67.3%of S.epidermidis strains were no slime forming.There was relation between PIA and antibiotics susceptibility.(P〈0.01).10 strains were icaA positive 11 strains were icaD positive by PCR analysis.There was not relation between icaA and icaD.(P0.05).7 strains were icaR positive,It was not relation betwen icaA or and biofilm formation icaD.(P0.05).CONCLUSIONS Resistant of S.epidermidis to Oxacillin in our hospital is mainly related to PIA synthesis and biofilm formation.The co-expression of icaA and icaD plays an important role in biofilm formation and the expression ica R gene inhibits the synthesis of PIA.
