摘  要:目的:建立大鼠动脉粥样硬化(AS)模型,观察动脉粥样硬化早期病变的动态变化过程和参莲方的干预作用。方法:240只大鼠随机分为空白对照组、模型组、阳性药组(阿司匹林0.027 g.kg-1)、参莲方高、中、低剂量组(0.675,1.35,2.70 g.kg-1)。采用球囊损伤法+半量高脂饲料建立AS模型,并给予参莲方治疗。HE法观察血管病理学变化,采用酶比色法、清除法、硝酸还原酶法测定TC,TG,LDL-C,HDL-C,NO水平的变化,放免法观察ET,TXB2,6-keto-PGF1α,TNF-α,IL-6及IL-8水平的变化。结果:参莲方各剂量组和同阶段模型组相比血管损伤均有明显改善,内膜炎细胞浸润、中膜增厚和平滑肌增殖减轻,第8周末参莲方组无明显脂质条纹出现。参莲方各剂量组血清TG,NO,TXB2的含量明显降低(P<0.05);高剂量组可明显降低血清TNF-α,IL-6,IL-8水平(P<0.05)。结论:参莲方能够通过调节血脂,改善血管内皮功能,减少血液循环中炎症介质的释放,抑制AS病变的形成和发展。Objective: To establish the atherosclerosis models(AS) in rats and observe the change process of AS and Shenlian decoction’s pharmacological effect.Method: Two hundreds and forty rats were randomly divided into blank control group,model group,positive medicine group,the high-dose,middle dose and low-dose Shenlian decoction groups.Vascular pathological changes were observed by the methods of HE stain.The levels of TC,TG,LDL-C,HDL-C,NO in serum were determined respectively by enzyme colorimetric,clear and nitrate reductase methods.ET,TXB2,6-keto-PGF1α,TNF-α,IL-6 and IL-8 levels in serum were detected by radioimminoassay.Result: Compared with the model group,vascular lesion of different doses of Shenlian decoction groups were significantly improved,the TG,NO,TXB2 levels in serum were significantly lower(P〈0.05),while high-dose Shenlian decoction can significantly reduce TNF-α,IL-6,IL-8 levels in serum(P〈0.05).Conclusion: Shenlian decoction can inhibit the formation and development of AS by regulating blood lipids,improving endothelial function and reducing inflammatory factors release.
