摘 要:目的比较冷冻切片琥珀酸脱氢酶(succinate dehydrogenase,SDH)酶组织化学染色法和石蜡切片HE染色法在评价高强度聚焦超声(HIFU)消融牛肝组织即刻效应中的价值。方法 HIFU消融牛肝组织后即刻,分别置液氮和4%多聚甲醛中,分别行冷冻切片SDH酶组织化学染色和石蜡切片HE染色。以未消融牛肝为对照。结果 HIFU消融后即刻,SDH酶组织化学染色显示消融区内SDH活性消失,HE染色显示细胞核形态及细胞结构无明显改变。2种方法检测的有效性差异显著(P<0.05)。结论冷冻切片SDH酶组织化学染色可作为评价HIFU消融即刻效应快速有效的方法。Objective To compare the value of enzyme histochemical staining of frozen section for succinate dehydrogenase (SDH) and H&E staining of paraffin section during evaluating the immediate effect of the bovine liver ablation by High Intensity Focused Ultrasound ( H1FU). Methods After HIFU ablation, the bovine liver pieces were placed respectively into the liquid nitrogen for enzyme hislochemical staining of frozen section for SDH and into the 4% paraformaldehyde for H&E staining of paraffin section. The bovine liver tissue without ablation was served as control group. Results After the HIFU ablation, immediately the bovine liver in enzyme histochemical staining showed that SDH activity was lost in ablated region. In contrast, there were no significant changes in nuclei and cellular architectures displayed by H&E staining. There was significant difference in effectiveness between the two groups(P〈0.05). Conclusions Frozen section for SDH enzyme histochemical staining could be considered as a rapid and effective means to evaluate the immediate effect of HIFU ablation.
