摘  要:目的:通过藕节炒炭前后提取物、藕节不同提取部位及有效成分对小鼠凝血时间的影响观察,验证藕节炒炭止血的科学性,确定藕节炭止血作用的有效部位,为探讨止血作用机制奠定基础。方法:采用小鼠凝血时间作为主要(毛细玻管法)观察指标,分别对藕节炒炭前后受试液、藕节不同提取部位及有效成分进行了测定,18~22g昆明种小鼠,按体重、性别均衡、随机分组,分别给予相应受试药物。每天给药1次,连续10天。于最后一次给药后60min,用毛细玻璃管自小鼠目外眦采血,自采血开始计时,观察自采血至出现血丝的时间作为凝血时间,比较受试药物组凝血时间的变化。结果:通过凝血时间试验,对生藕节提取物、藕节炭提取物进行了凝血时间影响的筛选,确定藕节炒炭后止血作用优于生藕节的止血作用。在前次实验基础上对藕节石油醚提取部位、醋酸乙酯提取部位、水提取部位进行了凝血时间影响的筛选,确定藕节止血作用的有效部位为醋酸乙酯部位和水提取部位。进一步观察藕节炭醋酸乙酯提取物、3-表白桦脂酸对小鼠凝血时间的影响,提示两组受试药均能缩短小鼠凝血时间。结论:藕节炒炭后止血作用加强,醋酸乙脂提取部位、水提取部位为藕节炭止血作用的主要活性部位3,-表白桦脂酸为藕节炭止血作用的有效成分之一。Objective:To Oujie fried carbon before and after the extract,Ou Jie and effective ingredients of different extracts of mouse coagulation time of observation,verification Oujie carbon bleeding scientific speculation to determine the role of an effective hemostatic Oujie carbon site,to explore the mechanism of bleeding basis. Methods:The blood coagulation time in mice as the primary (glass capillary tube method) to observe targets,respectively,by the test solution before and after carbon Oujie fried,Ou Jie and effective ingredients of different extracts were determined,18 ~ 22g Kunming mice,according to body weight,gender balance, randomized subjects were given the appropriate drugs. Administered once a day for 10 days. After administration in the last 60min,with the capillary tube outer canthal childhood Shumu Blood samples were collected from the blood start running,observe from the blood to appear bloodshot time as the clotting time compared drug group subjects clotting time changes. Results:The blood coagulation time tests on raw Oujie extract,extract carbon Oujie clotting time of the impact of screening,to determine the role of Oujie fried charcoal to stop bleeding after the bleeding is superior to the role of raw Oujie. In the previous experiments on the basis of Oujie petroleum ether extracts,ethyl acetate extracts,water extracts had the impact of screening coagulation time to determine the role of an effective hemostatic Oujie site location for the ethyl acetate and water extracts. Further observation Oujie charcoal ethyl acetate extract,3-Table betulinic acid effects on clotting time in mice,suggesting that two groups of subjects are able to shorten the drug coagulation time in mice. Conclusion:Oujie fried charcoal to stop bleeding after the greater role of ethyl acetate extracts,water extracts of the role of major bleeding Oujie carbon active sites,3-Table betulinic acid as a hemostatic effect of carbon Ou Jie,one of the active ingredients.
