升清降糖合剂对STZ诱导RINm5F细胞损伤的保护作用

摘　　要：目的:探讨升清降糖合剂对RINm5F细胞的保护作用,为临床治疗糖尿病提供实验依据。方法:以STZ(链脲佐菌素)诱导建立胰岛细胞损伤模型;采用半仿生提取法提取升清降糖合剂有效成分(简称SQ);以不同浓度(100、200、400μg.mL-1按生药折算)的SQ作用于正常组和模型组细胞,以CCK-8法检测细胞生存活性并结合硝酸酶还原法测定细胞上清中NO含量。研究结果:经CCK-8法检测确定建立模型条件为0.8mmol/LSTZ干预RINm5F细胞24h;400μg/mL药物组能明显提高细胞活性(P<0.01),并减少细胞培养液中NO含量(P<0.01)。研究结论:升清降糖合剂对STZ导致胰岛细胞损伤有保护作用。Objective：To investigate the protective effect of Sheng Qing Jiang Tang He Ji on RINm5F.Methods：We obtained Sheng Qing Jiang Tang He Ji（SQ） active ingredients using semi-bionic extraction method.To establish an injury model of RINm5F cells with streptozotocin（STZ）.Then applying CCK-8 colorimetric method to observe the changes of cell survival activity and nitrate reductase method to study the effect of medicine on NO content of cultured cell supernate fluid.Results： RINm5F’s vitality reduced to IC50 when explored in 0.8mmol/L STZ for 24h.In cell survival activity study（CCK-8）,the OD value of Sheng Qing Jiang Tang He Ji 400μg/mL is significantly higher than model group（P〈0.01）.To compare with model group,Sheng Qing Jiang Tang He Ji 400μg/mL can decrease NO contents（P〈0.01）.Conclusion：The protective function of Sheng Qing Jiang Tang He Ji on RINm5F is probably by increasing cell survival activity and reducing NO contents.
