摘  要:BLM解旋酶是RecQ家族DNA解旋酶中的一个重要成员,在维持染色体的稳定性中具有重要的作用,其突变会导致Bloom综合症。Bloom综合症是一种罕见隐性常染色体遗传疾病,患者易患各种类型癌症。本研究利用紫外吸收光谱法研究了pH、反应时间、NaCl浓度对BLM解旋酶的稳定性和ATP酶活性的影响。结果显示,pH会影响BLM解旋酶的构象和ATP酶活性,碱性环境对其构象和ATP酶活性影响比酸性环境大。酶在稀释的过程中对其构象和ATP酶活性有影响。NaCl浓度小于500mmol/L时,对酶构象和ATP酶活性影响不大;当NaCl浓度大于500mmol/L时,酶的构象和ATP酶活性均受到影响。这些研究结果将为开展小分子药物与BLM解旋酶的相互作用研究提供相关资料。BLM helicase is an important member of the RecQ family DNA helicase and plays an important role in the maintenance of genetic stability,and its mutation could lead to the Bloom’s syndrome.Bloom’s syndrome(BS) is a rare,autosomal recessive genetic disorder,and patients is easy to sick the most types of cancers.The effects of pH,reaction time,and concentrations of NaCl on the stability and ATP activity of BLM helicase were investigated by ultraviolet absorption spectroscopy.The results indicated that the conformation and ATP activity of BLM helicase were affected by the pH,the effects of alkaline environment on which were greater impact than acid environment.The conformation and ATP activity of BLM helicase had a few of effects in dilution process.The NaCl solution had few effects on the conformation and ATP activity of BLM helicase when the concentrations were less than 500mmol/L,but the conformation and ATP activity were affected when the concentrations were more than 500mmol/L.These results may provide the associated materials to study the interaction of small-molecule drugs and BLM helicase.
