摘 要:考察了万古霉素的稳定性及其植入剂的体外药物释放动力学,比较了不同取样方法:顺序取样法、部分换液法和等时间间隔全换液取样法,对万古霉素释放行为的影响。结果表明,顺序取样法和部分换液取样法对体外释放测定结果影响显著,得到万古霉素回收率为26.2%和42.7%;而用等时间间隔全换液取样法得到万古霉素回收率为93.6%。因此,等时间间隔全换液取样法更适用于万古霉素植入剂的体外释放研究。The drug stability and release behavior of vancomycin from its implants in vitro was studied. The influences of three kinds of sampling methods, such as sequential sampling, partially replacement of phosphate buffer solution (PBS) sampling and total replacement of PBS sampling every 24 h on the results of in vitro release tests were investigated. The results showed that sequential sampling and partially replacement of PBS sampling had significant effects and the recoveries of vancomycin were 26.2 % and 42.7 %. The method of total replacement of PBS sampling every 24 h with the recovery of 93.6 % turned out to be more reliable for evaluating vancomycin in vitro release from its implants.
