摘 要:目的观察温经汤对寒凝血瘀模型大鼠卵巢血红素氧合酶1(HO-1)、血红素氧合酶2(HO-2)的影响,探讨温经汤治疗寒凝血瘀型妇科疾病的作用机制。方法按随机数字表法将40只SD雌性大鼠分为正常组、模型组、温经汤低剂量组(低剂量组)、温经汤高剂量组(高剂量组),每组10只。低剂量组、高剂量组于造模的第1天开始,分别按14.66、29.32g/(kg.d)-1给予温经汤灌胃,每日1次,连用2周。其余两组给予同体积的蒸馏水。采用血红蛋白结合及联二亚硫酸盐还原法测定血浆一氧化碳(CO)活性;采用免疫组织化学SP法检测卵巢颗粒细胞和黄体细胞HO-1、HO-2蛋白表达;采用原位杂交法检测卵巢颗粒细胞和黄体细胞HO-1、HO-2 mRNA的表达。结果高剂量组、低剂量组大鼠血浆碳氧血红蛋白(COHb)活性较模型组明显增强(P<0.05或P<0.01);卵巢颗粒细胞、黄体细胞HO-1、HO-2蛋白表达,HO-1 mRNA、HO-2 mRNA表达加强(P<0.05或P<0.01)。结论温经汤治疗寒凝血瘀型妇科疾病的作用可能与调节HO-1、HO-2水平有关。Objective To observe the effect of Wenjing Decoction (Decoction for warming meridians) on the expression of heme oxygenase 1(HO-1) and heme oxygenase 2(HO-2) in the ovary of model rats with blood stasis due to cold accumulation,and to explore its therapeutic mechanism in treating gynecological diseases.Methods Forty SD female rats were randomized into normal group,model group,and Wenjing Decoction low-dose and high-dose groups,10 in each.Wenjing Decoction low-dose and high-dose groups were administered Wenjing Decoction by gavage 14.66 or 29.32g/(kg·d)-1 respectively,once a day,for 2 weeks in succession.The other two groups were given the same volume distilled water.The plasma carbonic oxide(CO) activity was determined with the method of hemoglobin binding and dithionite reduction,the expression of HO-1 and HO-2 in ovarian granular and luteal cells was determined with the immunohistochemical SP method,and the expression of HO-1 mRNA and HO-2 mRNA in ovarian granular and luteal cells was determined with the in situ hybridization method.Results Compared with the model group,the activity of plasma COHb in the Wenjing Decoction low-dose and high-dose groups was significantly increased(P0.05 or P0.01),while the expressions of HO-1,HO-2,and HO-1 mRNA and HO-2 mRNA in ovarian granular and luteal cells were dcreased(P0.01 or P0.05).Conclusion The therapeutic effect of Wenjing Decoction on gynecological disease due to cold accumulation and blood stasis was related to regulating the expression of HO1 and HO-2.
