摘 要:目的:通过检测急性放射损伤小鼠骨髓组织中血管内皮细胞生长因子(VEGF)、粒细胞集落刺激因子(G-CSF)的表达及骨髓细胞超微结构的变化,探讨槐白皮对骨髓造血细胞和造血微环境的影响。方法:实验随机分为5组,即正常、照射对照和槐白皮高、中、低剂量(0.8、0.4、0.2mL·kg-1)组。采用免疫细胞化学SABC染色法检测放射损伤小鼠骨髓组织切片中VEGF和G-CSF的表达水平,透射电镜检测骨髓细胞超微结构的变化。结果:槐白皮高、中、低剂量组小鼠骨髓组织中VEGF的表达均显著高于照射对照组(P<0.01);槐白皮中剂量组小鼠骨髓组织中G-CSF表达显著高于照射对照组(P<0.01),但槐白皮高、低剂量组与照射对照组比较无显著性差异(P>0.05)。与正常组比较,槐白皮中剂量组小鼠骨髓组织中VEGF、G-CSF表达无显著性差异(P>0.05),槐白皮高、低剂量组表达水平低于正常组(P<0.05)。骨髓细胞超微结构显示,槐白皮治疗后其细胞排列整密,染色质均匀,无核固缩,粗面内质网较丰富,轻度扩张。结论:槐白皮能改善急性放射损伤小鼠骨髓血管微环境,增强骨髓造血细胞的恢复。OBJECTIVE:To determine the expression of vascular endothelial growth factor(VEGF),granulocyte colony-stimulating factor(G-CSF) and ultra-structure of bone marrow in mice with acute radiation injury,in order to evaluate the effect of Sphora japonica on the hematopoietic microenvironment and hematopoietic cells of bone marrow.METHODS:40 mice were randomly divided into 5 groups:normal group,irradiation control group,S.japonica high-dose,medium-dose and low-dose groups(0.8 mL·kg-1,0.4 mL·kg-1 and 0.2 mL·kg-1).The expressions of VEGF and G-CSF in bone marrow tissue were determined by immunocytochemistry SABC staining.Transmission electron microscope was used to observe the change of ultra-structure of bone marrow cells.RESULTS:The expression of VEGF in bone marrow tissue in S.japonica high-dose,medium-dose and low-dose groups was significantly higher than in irradiation control group(P0.05).The expression of G-CSF in S.japonica medium-dose group was significantly higher than in irradiation control group(P0.01),but there was no significant difference in S.japonica high-dose and low-dose groups(P0.05).There was no significant different in the expression of VEGF and G-CSF between S.japonica medium-dose group and normal group(P0.05),but the expression of VEGF and G-CSF in S.japonica high-dose and low-dose groups was lower than normal group(P0.05).The ultra-structure of bone marrow showed bone marrow cells arranged regularly and closely,chromatin was uniform,pyknotic nuclear was not found,rough endoplasmic reticulum was abundant but expanded mildly.CONCLUSIONS:S.japonica can improve the hematopoietic microenvironment in mice with acute radiation injury and promote the growth of hematopoietic cells.
