Optimization of the Preparation Technology of Amomum villous Oil-hydroxypropyl-β-Cyclodextrin Inclusion Complex by Orthogonal Experiment

摘  要:目的:研究羟丙基-β-环糊精(HP-β-CD)包合春砂仁挥发油的最佳工艺,并进行包合物鉴定。方法:以挥发油利用率、包合物含油率与最高含油率比值和包合物收率的综合得分为评价指标,对搅拌法、超声法和研磨法进行优选;以挥发油与HP-β-CD的投料比、HP-β-CD的浓度、包合时间、包合温度为考察因素,上述综合得分为评价指标,采用正交试验优选最佳包合工艺,并通过显微鉴别、差示扫描量热分析、紫外分光光度、气相色谱质谱联用等方法对包合前、后的包合物进行鉴别。结果:最佳包合方法为超声法,最佳包合工艺条件为挥发油与HP-β-CD的投料比为1:10(V:m),HP-β-CD的浓度为40%,包合时间80min,包合温度55℃。包合前、后成分未见明显变化。结论:春砂仁挥发油的HP-β-CD包合工艺切实可行,可为实际生产研究提供一定的试验依据。OBJECTIVE:To optimize the preparation technology of Amomum villous oil-hydroxypropyl-β-cyclodextrin(HP-β-CD)inclusion complex,and to identify the inclusion complex.METHODS:With the utilization ratio of volatile oil,ratio of oil content to the highest oil content and yield of inclusion complex as index,the stirring method,ultrasonic method and trituration method were optimized.The preparation technology of inclusion complex was optimized by orthogonal experiment taking the comprehensive evaluation score as index with ratio of volatile oil to HP-β-CD,concentration of HP-β-CD,inclusion time and inclusion temperature as factors.Microscopic identification method,DSC method,UV method and GC-MS method were used for the identification of inclusion complex.RESULTS:Ultrasonic method was the best preparation method,and the optimal preparation condition as following:the ratio of volatile oil to HP-β-CD was 1:10(V:m),the concentration of HP-β-CD was 40%,the time and temperature for inclusion was 80 min and 55℃.No obvious change was found before and after inclusion.CONCLUSION:The optimized preparation technology of A.villous oil-hydroxypropyl-β-cyclodextrin is feasible and serves as reference for the production and research.
