摘  要:目的:建立测定导赤丸中2种有效成分黄芩苷和大黄素的含量的方法。方法:采用高效液相色谱法。色谱柱为Chroma-trex C18柱;测定黄芩苷的流动相为甲醇-水(加入磷酸调pH=3.5,43:57),检测波长为280nm,流速为1.0mL·min-1,柱温为30℃;测定大黄素的流动相为甲醇-水(加入冰醋酸调pH=2.0,80:20),检测波长为245nm,流速为1.0mL·min-1,柱温为25℃。结果:黄芩苷和大黄素的检测浓度分别在0.626~1.044mg·mL-1(r=0.9996)和5~25μg·mL-1(r=0.9998)范围内线性关系良好;平均回收率(n=9)分别为100.18%(RSD=0.75%)和99.30%(RSD=1.5%)。结论:本方法操作简便、准确度高、专属性强,适用于导赤丸的质量控制。OBJECTIVE:To establish the method for the content determination of baicalin and emodin in Daochi pills.METHODS:HPLC was adopted with chromatrex C18 as column.The mobile phase for baicalin determination consisted of methanol-water-phosphoric acid(43:57) with detection wavelength at 280 nm.The flow rate was set at 1.0 mL·min-1 and the column temperature was 30 ℃.The mobile phase for emodin determination was methanol-water-glacial acetic acid(80:20) with detection wavelength at 245 nm.The flow rate was 1.0 mL·min-1 and the column temperature was 25 ℃.RESULTS:There was good linearity between the peak area and concentration within the ranges of 0.626~1.044 mg·mL-1(r=0.999 6) for baicalin and 5~25 μg·mL-1(r=0.999 8) for emodin,respectively.The average recoveries of baicalin and emodin were 100.18%(RSD=0.75%)and 99.30%(RSD=1.5%),respectively(n=9).CONCLUSION:The method is simple,accurate and specific for quality control of Daochi pills.
