摘  要:目的筛选益气健脾中药黄芪和白术促进小肠上皮细胞(IEC-6)迁移的有效提取部位。方法以系统溶剂分离法从黄芪、白术药材分别得到糖复合物、正丁醇萃取物、醇沉上清液以及大孔树脂吸附后70%乙醇洗脱部位作受试药。IEC-6细胞接种于6孔板并培养24 h,划痕法造细胞迁移模型,划痕后加入各受试药,24 h后观察细胞迁移情况;并进一步观察具有促进细胞迁移的有效部位对二氟甲基鸟氨酸(DFMO)所致细胞迁移抑制的影响。结果 50 mg.L-1的黄芪糖复合物或白术糖复合物能促进细胞迁移(P<0.01);黄芪、白术的正丁醇萃取物、醇沉上清液、大孔树脂吸附后醇洗脱部位对细胞迁移均无明显影响;2.5 mmol.L-1的DFMO能抑制细胞迁移(P<0.01),加入DFMO同时加入100 mg.L-1黄芪糖复合物或200 mg.L-1白术糖复合物能使细胞迁移恢复至接近正常水平。结论黄芪糖复合物和白术糖复合物能促进IEC-6细胞迁移,还能使DFMO所致的细胞迁移抑制恢复至接近正常水平。Objective To screen the effective fraction of Radix Astragali and Rhizoma Atractylodis Macrocephalae, which have the actions of strengthening spleen and replenishing Qi, for promoting intestine epithelial cell 6 (IEC-6)migration. Methods The testing agents included polysaccharides, N-butanol extracts and supernatant of ethanol precipitation from Radix Astragali and Rhizoma Atractylodis Macrocephalae prepared by systematic solvent extraction firstly, and then with 70 % ethanol eluent after D-101 macroporous resin adsorption. Cells were implanted in 6-well plates. Cell migration model was induced by scratch method in each well after cuhuring for 24 h, and then the CDMEN contained testing agents was added in. Twenty-four hours later, the migrating cells were counted to screen the effective fraction for promoting cell migration. The effects of the extracts on promoting cell migration were observed by giving a migration inhibiter of -difluoromethylornithine (DFMO). Results Polysaccharides from Radix Astragali(50 mg·L^-1)and Rhizoma Atractylodis Macrocephalae(50 mg·L^-1) both promoted IEC-6 cell migration(P 〈 0.01 compared with the blank group). N-butanol extract, ethanol supernatant prepared by systematic solvent extraction, and 70% ethanol eluent prepared by D-101 macroporous resin adsorption from Radix Astragali and Rhizoma Atractylodis Macrocephalae had no significant affect on the cell migration. DFMO (2.5 mmol·L^-1)inhibited the cell migration(P〈 0.01 compared with the blank group). The polysaccharides from Radix Astragali( 100 mg·L^-1)and Rhizoma Atractylodis Macrocephalae(200 mg·L^-1)reversed the DFMO-induced cell migration inhibition to normal level(P 〈 0.01and P 〈 0.05 compared with the model group; P〉 0.05 compared with the blank group). Conclusion The polysaccharides from Radix Astragali and Rhizoma Atractylodis Macrocephalae p can promote IEC-6 cell migration, and reverse the DFMO-induced cell migration inhibition to normal levels.
