摘  要:目的比较源于肝癌患者与健康人的细胞因子诱导杀伤(CIK)细胞的生物学特性及抗癌活性。方法提取肝癌患者和健康人的外周血单个核细胞,常规诱导培养CIK细胞,动态观察其增殖活性、细胞表型及细胞毒活性。选BALB/c裸鼠SMMC-7721肝癌模型,接种肿瘤细胞10d后,选择荷瘤大小相似的裸鼠30只,腹腔注射化疗药物后平分成A组(单独化疗组)、B组(健康人CIK治疗组)和C组(肝癌患者CIK治疗组),观察肿瘤大小的变化。结果两种来源CIK细胞的增殖活性、细胞表型及细胞毒活性均差异无显著性,体外抗癌活性相似,均可提高化疗的效果。B组和C组治疗后肿瘤受抑制的程度差异无显著性,但均明显大于A组。结论源于肝癌患者CIK细胞与源于健康人的CIK细胞具有相似的生物学特性及抗癌活性。[ Objective ] To cmnpare the biological characteristics and anti-cancer activities of human cytokineinduced killer (CIK) cells from patients with hepatocellular carcinoma and healthy donors. [Method] Peripheral blood mononuclear cells from patients with hepatocellular carcinoma and healthy blood donors were incubated in vitro and induced into CIK cells in the presence of various cytokines, such as interferon-gamma, interleukin-1, IL-2, and monoelonal antibody against CD3. The phenotype and proliferation activity of CIK cells were identified by flow eytometric analysis. The cytotoxicity of CIK cells in vitro was determined by MTF assay. The anti-tumor activities of CIK cells were evaluated after chemotherapy in BALB/c nude mice with SMMC-7721 liver cancer.[Result] CIK cells from both groups didn't show obvious differences in phenotype, proliferation activity, cytotoxicity, and auxiliary therapeutic effects. CIK ceils enhanced the effects of the chemotherapy in liver cancer model. [ Conclusion ] The biological characteristics and anfitumor activities of CIK cells from patients with hepatocellular carcinoma were almost the same as those from healthy donors.
