摘 要:利用ISSR和SRAP分子标记对11份不同地理居群野生菊的遗传多样性进行了分析。结果发现:25个ISSR引物共扩增到203条扩增带,其中有187条呈多态性,多态性比率平均为91.9%;29对SRAP引物组合共扩增到446条扩增带,其中有435条呈多态性,多态性比率平均为97.5%;11个居群间的Ne′is遗传距离分布在0.088 7-0.615 5之间。聚类分析表明,紫花野菊、野路菊及其变种能很好地区分开来,但是野生菊资源的遗传变异与地理分布相关性似乎并不明显。Genetic diversity among eleven populations of wild species of Dendranthema was analyzed by the molecular markers of ISSR and SRAP.The results showed that ISSR fingerprinting amplified by 25 ISSR primers revealed a total number of 203 unambiguous bands of which 187 ones were polymorphic and the polymorphism frequency was 91.9%;and SRAP fingerprinting amplified by 29 SRAP primer pairs revealed a total number of 446 unambiguous bands,of which 435 ones were polymorphic and the polymorphism frequency was 97.5%,which indicated a rich genetic diversity of wild species of Dendranthema;The genetic distances ranged from 0.088 7 to 0.615 5.The UPGMA dendrogram suggested that there was no significant relationship between the geographical distribution and genetic diversity,and that it could separate D.japonense(or it′s varieties)from D.zawadskii(or it′s varieties).
