Effect of transgenic hybrid poplar 741 with two insect-resistant genes on primary detoxification enzyme and acetylcholinesterase of target pests

摘  要:用不同系号转基因741杨叶片饲养杨扇舟蛾、舞毒蛾和美国白蛾4~6龄幼虫,测定其中肠重要的两种解毒酶和乙酰胆碱酯酶比活力,研究转基因741杨对靶标害虫生理代谢的影响。结果表明,转基因741杨显著抑制了羧酸酯酶、谷胱苷肽-S-转移酶和乙酰胆碱酯酶的比活力,其中PB11和PB29的抑制作用达到了极显著水平,与转基因741杨抗虫性的生测结果一致。转基因741杨对靶标害虫解毒酶和乙酰胆碱酯酶比活力的抑制作用是其抗虫性的生理机制之一。Fourth-sixth instar larva of Clostera anachoreta (Fabricius), Gypsy moth and American white moth were fed on various clones of transgenic 741 poplar leaves and the activities of two detoxification enzymes and acetylcholinesterase in midgut were investigated to study the effect of transgenic 741 poplars on the physiological metabolism of the target pests. The results showed that the activities of carboxyleste- rase, glutathione-S-transferase and acetylcholinesterase were inhibited by transgenic 741 poplars, and the inhibition of PB11 and PB29 reached significantly different level. The results were consistent with that of insect-resistant bioassay. The inhibition of transgenic 741 poplars on the activities of detoxification enzymes and acetylcholinesterase of target pests is one of the physiological mechanisms of their insect-resistance.
