摘 要:采用改良的CTAB法提取番茄4个品种及7份品种亲本的基因组DNA,通过AFLP-PCR分子标记方法进行DNA指纹分析,构建亲本及品种的指纹图谱。从20对引物中筛选出9对重复性好,多态性丰富的AFLP引物来构建番茄指纹图谱,7份亲本材料扩增的多态性位点从10~27条不等,平均每份材料17.56条,多态性位点比例为41.25%。通过田间纯度的符合率测定,亲本纯度的符合率为100%,杂种一代为98.7%;4个番茄品种及7份亲本材料的遗传相似系数分别在0.48~0.71和0.62~0.76之间,利用UPGMA法进行聚类分析,从分子水平对4个番茄品种及7份亲本材料的遗传关系进行分析,为番茄杂交育种和品种鉴定提供理论依据。Total genomic DNA of four tomato varieties and their parents was extracted by using CTAB method. DNA fingerprinting maps of four tomato varieties and their parents were established byAFLP-PCR method. The results showed that nine primers from 20 primers were selected. The amplified polymorphic bands of the parents were from 10 to 27, and the average bands was 17.56, the percentage of polymorphic bands was 41.25%. The purity of seven parents was 100%, and that of the tomato varieties was 98.7%. The genetic similarity coefficients of four varieties and their parents were from 0.48 to 0.71 and from 0.62 to 0.76. The cluster analysis was conducted and the genetic similarity coefficient was calculated by using UPGMA method. This study will provide the theory basis for tomato cross breeding and cultivars identification.
