摘 要:瓜类果斑病菌(Acidovorax avenae subsp.citrulli,Aac)是瓜类作物上重要的病原细菌,为我国进境植物检疫性有害生物。胶体金免疫层析试纸条方便快捷,应用广泛。该方法使用不当会出现假阳性问题,仅适用于病原菌的初筛检测。本研究将Aac胶体金免疫层析方法(GICA)与PCR技术相结合,建立了GICA-PCR检测方法。检测结果表明,该方法在蛋白与核酸2个层面上从发病西瓜叶片上检测到瓜类果斑病菌,有效解决了试纸条检测的假阳性问题,提高了瓜类果斑病菌检测的准确性,值得推广应用。Acidovorax avenae subsp, citrulli ( Aac), the cause agent of Bacterial fruit blotch in Watermelon, is a plant quarantine pest in China. Gold Immunochromatographic Assay (GICA) Strip is widely used for bacteria di- agnosis because of its performing convenience and directly easy visual judging. However, the strip is only suitable for bacteria preliminary test since false positive result maybe appeared. In this research, we developed one GICA - PCR method combined the GICA and PCR assay. The test line of the GICA strip wascut and soaked in 20t^L water as the template of PCR reaction. From the bacteria culture and infected watermelon leaves Aac can be detected by using GICA -PCR. When the test line is not clearly seen PCR result is also test positive. In this assay GICA is based on the protein and PCR method is nucleic acid, then this method can effectively reduce the false positive of the strip and can improve the detection sensitivity. Therefore, the GICA -PCR method is worth to be applied for the detection of Bacterial Fruit Blotch.
