摘 要:目的:观察芪红颗粒对慢性心力衰竭大鼠血清脑钠肽(BNP)和血清基质金属蛋白酶-9(MMP-9)的影响,探讨其防治心力衰竭的机理。方法:采用皮下注射异丙基肾上腺素的方法复制心力衰竭大鼠模型。6周行超声心动图测定证明模型成功,将成模大鼠随机分为模型组、芪红颗粒低剂量组(QHD)、芪红颗粒中剂量组(QHZ)芪红颗粒高剂量组(QHG)和地高辛组,分别灌胃4周后再次行超声心动图测定,利用酶联免疫法检测血清BNP、MMP-9水平,观察各组大鼠心肌病理形态学特征。结果:异丙基肾上腺素致慢性心力衰竭大鼠心功能下降,BNP、MMP-9明显升高、大鼠心肌病理形态学明显改变;治疗4周,各治疗组均有不同程度改善,其中芪红颗粒中、高剂量组作用优于地高辛组,统计有显著性差异(P<0.01)。结论:芪红颗粒能显著改善心功能、减轻心力衰竭症状,逆转心室重构,其作用可能与抑制心力衰竭大鼠血清BNP、MMP-9表达有关。Objective:To observe the effect of Qihong Granule on brain natriuretic peptide(BNP) and serum matrix metalloproteinase-9(MMP-9) in rats with chronic heart failure and to explore the mechanism of the prevention and treatment of heart failure.Methods: The method of subcutaneous injection of isoproterenol was taken to make model rat with heart failure.The echocardiography 6 weeks later proved the successful model,then the model rats were randomly divided into model group,low dose group of Qihong Granule(QHD),medium dose group of Qihong Granule(QHZ),high dose group of Qihong Granule(QHG) and digoxin group.They were measured again with echocardiography 4 weeks after the gastric perfusion.The enzyme-linked immunosorbent was used to examine the serum BNP,MMP-9 levels and to observe the morphological features of cardiac pathology.Results: Isoproterenol induced the decline of cardiac function of rats with chronic heart failure,increased BNP and MMP-9 significantly and the myocardial pathology changed significantly.Four weeks after the treatment,all treatment groups improved in different degrees,in which the effects of medium dose group of Qihong Granule(QHZ) and high dose group of Qihong Granule were superior to that of digoxin group,there was significant statistical difference(P0.01).Conclusion: Qihong Granule can significantly improve the heart function,reduce the symptoms of heart failure,reverse the ventricular remodeling,whose effect on heart failure may be related to the inhibition of serum BNP,MMP-9 expression.
