摘 要:[目的]探讨谷氨酸信号通路在黑素细胞中的作用机制。[方法]培养人原代黑素细胞。采用多巴染色法及透射电镜鉴定细胞;以Western Blot方法测定人原代黑素细胞内NMDAR2A的表达。采用扫描电镜观察谷氨酸受体激动剂N-Methyl-D-aspartic acid(NMDA),Quisqualate(Q-LA)和非竞争性拮抗剂MK-801作用下黑素细胞树突形态的变化。[结果]Western Blot实验发现人原代黑素细胞内有NMDAR2A蛋白的表达。扫描电镜观察发现谷氨酸受体激动剂NMDA及Q-LA作用后细胞树突长度明显变短,树突末端明显变宽,二、三级树突增加;谷氨酸受体拮抗剂MK801作用后细胞树突末端明显变窄,二、三级树突减少。[结论]除神经系统外,黑素细胞内存在谷氨酸信号通路,且该信号通路与细胞形态有关。[Objective] To investigate the role of glutamate pathway in melanocytes.[Methods] Human melanocytes were cultured primarily.Melanocytes were identificated by DOPA staining and scanning electron microscope;NMDAR2A expression were measured by Western Blot.The morphological change of cells after treatment with non-competitive NMDAR antagonist MK801,agonist N-Methyl-D-aspartic acid(NMDA),and gulamate receptor agonist of quisqualate(Q-LA).[Results] NMDAR2A protein was found in primary human melanocytes with scanning electron microscope.The dendrites of melanocytes became longer and the end of dendrites were flatter and presented with more second and third grade dendrites after treatment by NMDA and Q-LA,while the dendries became slender,second and third grade dendrites reduced after treatment of NMDAR2A antagonist MK-801.[Conclusion] There is glutamate signaling pathway,which could affect cellular morphology in melanocytes.
