摘 要:目的 探讨TGF-β/Smad信号转导通路与Wnt/β-catenin信号转导通路在病理性瘢痕发病机制中的作用及相互关系.方法 随机选取瘢痕疙瘩(K组)、增生性瘢痕(H组)和正常皮肤组织(N组)各3例,进行原代成纤维细胞培养,用不同浓度的TGF-β1处理细胞,应用免疫组化技术检测β-catenin在各组中的表达变化.结果 β-catenin在各组中的表达水平随TGF-β1浓度变化,呈先递增后递减趋势,三者β-catenin表达变化幅度为K组<H组<N组,K组成纤维细胞与H组成纤维细胞相比,差异无统计学意义(P>0.05),而N组成纤维细胞与K组、H组成纤维细胞相比,差异有统计学意义(P<0.01).结论 TGF-β/Smad信号转导通路与Wnt/β-catenin信号转导通路联合作用,导致病理性瘢痕形成.Objective To explore the interaction and interrelationship between the TGF-β/Smad signal pathway and Wnt/β-catenin signal pathway in pathogenesis of pathological scar. Methods Three cases of keloid ( K group), 3 of hyperplastic scar ( H group) and 3 of normal skin ( N group )were selected randomly, and then the fibroblasts taken from the 3 groups were performed the primary culture at different TGF-β1 concentrations and the expression of β-catenin was detected by immunohistochemisty technique. Results The expression of β-catenin in each group increased firstly and then decreased with the concentration changes of TGF-β1. The expression changes of the β-catenin in the three group were K group 〈 H group 〈 N group ,and there was no statistical significance in the variance between K group and H group( P 〉 0.05 ). There was substantial statistical significance in the variance between N group and K group, H group( P 〈0.01 ). Conclusion The unification of GF-β/Smad signal pathway and Wnt/β-eatenin signal pathway is an important factor in the formation of pathological scar.
