阿托伐他汀对糖尿病心肌病大鼠心肌细胞凋亡的影响

摘　　要：目的探讨阿托伐他汀对链脲佐霉素（STZ）制备的糖尿病心肌病（DCM）大鼠心肌细胞凋亡的影响。方法建立STZ诱导DCM大鼠模型，将实验大鼠分为对照（CON）组、阿托伐他汀预处理（Flu1）组、阿托伐他汀后处理（Flu2）组、糖尿病心肌病（DC）组。16周末处死，观察左室收缩功能（LVSP＋dp／dlmax）及舒张功能（LVEDP—ap／dtmax）；心肌细胞的凋亡百分数（TUNEL法）；心肌细胞超微结构的改变（电镜）。结果DC组与CON组相比，左室收缩、舒张功能明显降低，心肌组织中存在明显的细胞凋亡现象；与DC组相比，Flu1组、Flu2组左室收缩、舒张功能明显改善，心肌细胞凋亡指数下调；Flu2组与Flu1组相比，左室收缩、舒张及心肌细胞凋亡指数未见统计学差异。结论阿托伐他汀可有效降低糖尿病心肌病大鼠心肌细胞凋亡；单纯CRP升高而血糖正常的大鼠预防性给予阿托伐他汀与血糖升高时给予阿托伐他汀相比，两者对大鼠心肌细胞的保护作用未见统计学差异.Objective To investigate the effect of atorvastatin on apoptosis of cardiacmyocytes in rats with streptozocin（ STZ）-induced diabetic cardiomyopathy. Methods Wistar rats with type 2 diabetic cardiomyopathy model were induced with STZ. The rats were divided into saline control group（CON ） , atorvastatin prophylactic treatment group （ Flu1 ） ,atorvastatin after treatment group（Flu2 ） and diabetic cardiomyopathy group（DC）. At the end of 16th week,the left ventricular systolic pressure（ LVSP ＋ dp/dt ） and left ventricular end-diastolic pressure（ LVEDP -dp/dt ） were observed. Cadiac apoptosis was detected by terminal deoxynucleotidy transferase mediated dUTP-biotin nick end labelling （TUNEL） assay. Myocardial ultrastructure was examined by transmission electron microscopy（TEM）. Results Compared with CON,LVSP ＋ dp/dt LVEDP -dp/dt in DC LVSP, ＋ dp/dt LVEDP,-dp/dt in Flu1 and Flu2 were group were lower,but CAI was higher;Compared with DC, higher but CAI was lower;The difference of LVSP, ＋ dp/ dtmax., LVEDP,-dp/dt , CAI in Flu1 and Flu2 were not significant. Conclusion Atorvastatin can effectively inhibit the process of apoptosis of cardiacmyocytes in rats with STZ-induced diabetic cardiomyopathy. There was no differenee between atorvastatin prophylactie treatment on the rats with high CRP level and normol blood sugar and atorvastatin on the rats with hyperglycemia in the cardiacmyocytes protection of rats.
