摘 要:目的分析尖锐湿疣与HPV感染的关系,对比免疫组化和基因芯片对宫颈尖锐湿疣石蜡组织标本HPV检测的优缺点。方法应用基因芯片技术对29例宫颈尖锐湿疣组织进行HPV分型检测,并与免疫组化方法检测的HPV蛋白表达率对比。结果基因芯片技术HPV检出率为93.1%(27/29),明显高于免疫组化HPV检测阳性率69.0%(P<0.05)。基因芯片发现检出亚型主要为HPV6(9/29)和HPV11(20/29)。其中单一感染为81.5%(22/27),多重感染18.5%(5/27);共出现了1例单纯高危型HPV52感染和4例高低危型HPV复合感染(HPV65、6;HPV6、11、52;HPV6、11、58;HPV11、43、51、66)。结论宫颈尖锐湿疣与HPV6和HPV11感染密切相关。对石蜡标本HPV的检测,基因芯片技术与免疫组化相比,具有敏感性更高、特异性更强的特点。Objective To study the relationship between condyloma acuminate(CA) and human papilloma virus(HPV),and to compare the immunohistochemistry(IHC) and DNA chip technology for HPV detection in formalin-fixed paraffin-embedded(FFPE) specimens of CA. Methods IHC and DNA chip technology were employed to assess HPV protein expression and the HPV subtypes in 29 cases of FFPE CA samples,respectively. Results The HPV positive rate detected with DNA chip technology was 93.1%,which was significantly higher than that with IHC(69.0%) in the CA tissues(P0.05).HPV6(9/29)and HPV11(20/29)were found to be the most common subtypes of viruses.There were 22 cases of single infection in all 27 HPV positive cases,with 5 cases of multiple infection.Only 1 case of single infection with high-risk subtype(HPV52) and 4 cases of co-infection with high-risk and low-risk subtypes(HPV6,56;HPV6,11,52;HPV6,11,58;HPV11,43,51,66) were detected. Conclusion CA is closely related to HPV6 and HPV11 infection.DNA chip technology has higher sensitivity and specificity in detecting HPV infection in FFPE specimens,comparing with IHC.Furthermore,23 HPV subtypes can be typed with DNA chip technology.
