益气活血壮骨方对去势大鼠骨吸收与骨形成的影响

摘　　要：目的:探讨益气活血壮骨方对去势大鼠骨吸收与骨形成的影响。方法:将45只大鼠随机分为假手术组(A组)、模型组(B组)、益气活血壮骨方组(C组)、乐力胶囊组(D组)和骨松宝组(E组),每组9只。假手术组仅开腹缝合,其余4组做卵巢切除术。术后4周,A、B组大鼠以生理盐水灌胃(10 mL.kg-1),C组大鼠以益气活血壮骨方灌胃(9.6 g.kg-1),D组大鼠以乐力胶囊灌胃(0.15 g.kg-1),E组大鼠以骨松宝灌胃(8 g.kg-1),每天1次,连续给药12周。12周后处死大鼠,腹主动脉取血测定大鼠骨密度、骨矿含量、白细胞介素6、骨钙素及碱性磷酸酶。结果:益气活血壮骨方组骨密度及骨矿含量高于模型组,白细胞介素6、碱性磷酸酶和骨钙素含量低于模型组;益气活血壮骨方组白细胞介素6含量低于乐力胶囊组和骨松宝组;模型组骨密度、骨矿含量、白细胞介素6、骨钙素及碱性磷酸酶与其它组比较有统计学差异;假手术组与乐力胶囊组骨密度及骨矿含量、白细胞介素6和碱性磷酸酶比较有统计学差异。结论:益气活血壮骨方可抑制去势大鼠骨吸收,促进骨形成,从而阻止骨量丢失。益气活血壮骨方抑制破骨细胞分化增殖和骨吸收的作用强于乐力胶囊和骨松宝。Objective：To explore the effect of Yiqihuoxue Zhuanggu（YQHXZG） decoction（益气活血壮骨方） on bone resorption and bone formation in castrated rats.Methods：45 rats were randomly divided into sham-operated group（group A）,model group（group B）,YQHXZG decoction group（group C）,LELI（compound calcium amino acid chelate） capsules group（group D） and GUSONGBAO（a kind of traditional Chinese medicine） group（group E）,9 cases in each group.The cases in group A were administrated with sham-operation only,while those in other groups were administrated with oophorectomy.4 weeks after operation,rats in group A and group B were lavaged with physiological saline（10 mL·kg-1）,rats in group C were lavaged with YQHXZG decoction（9.6 g·kg-1）,rats in group D were lavaged with LELI capsules（0.15 g·kg-1）and rats in group E were lavaged with GUSONGBAO（8 g·kg-1）,once a day for consecutive 12 weeks.12 weeks later,all the rats were sacrificed,the bone mineral density（BMD）,bone mineral content（BMC）,interleukin-6（IL-6）,bone gla protein（BGP）and alkaline phosphatase（ALP）of the rats were measured.Results：BMD and BMC of group C were higher than those of group B,while the contents of IL-6,ALP and BGP of group C were lower than those of group B.The content of IL-6 of group C were lower than that of group D and group E.There was statistical difference in BMD,BMC,IL-6,BGP and ALP between group B and any other group.There was statistical difference in BMD,BMC,IL-6 and ALP between group A and group D.Conclusion：Through inhibiting bone resorption and promoting bone formation,Yiqihuoxue Zhuanggu decoction（益气活血壮骨方） performs well on preventing from bone loss for castrated rats,and it has a stronger effect of inhibiting osteoclast differentiation and proliferation and bone resorption than LELI capsules and GUSONGBAO.
