摘  要:利用显微镜观察长蛸Octopus variabilis Sasaki胚胎的发育过程,并采用生物化学方法对长蛸胚胎发育过程中可溶性蛋白的含量及组成变化进行分析和研究。结果显示,可溶性蛋白含量及组成变化与其胚胎发育的过程密切相关。在胚胎发育过程中,可溶性蛋白的含量呈先上升后下降的趋势。随着胚胎发育的进行,可溶性蛋白的组成由高分子质量向低分子质量的蛋白转化,相对分子质量为37kDa的谱带从黑珠期起缺失,相对分子质量为47 kDa的新谱带在器官形成期时出现,黑珠期和胚胎逆转期新谱带的表达量增加,相对分子质量为44.5 kDa的新谱带在黑珠期时出现,相对分子质量为41 kDa的新谱带在胚胎逆转期时出现。可溶性蛋白含量及组成的这些特征变化,可作为胚胎发育早期营养代谢的指标,指导人工育苗生产,提高胚胎孵化率。Octopus variabilis Sasaki belongs to Cephalopoda,Octopoda,Opisthoteuthidae,Octopus.It is a major economic species of seafood full of nutrition.O.variabilis is a traditional species of fishery with high economic value in China with large fishery amount.Females with high gonadal development were selected in sea area of Sanmenwan Bay in May 2009 before laying egg and hatching at 26~29 ℃ in hatching pond.In the present study,the embryonic development of O.variabilis was examined with microscopy and was divided into 6 phases.The 6 phases are zygote and cleavage phase,blastula and gastrula phase,organogenesis phase,hongzhu phase,heizhu phase and inversion phase.The composition and content of soluble proteins in the 6 phases of embryonic development were analyzed.The results showed that the composition and content of soluble proteins were in accordance with the embryonic development stage.The soluble proteins content rose in the zygote and cleavage phase,and it showed a downward trend with the progress of embryonic development.The soluble proteins content reached the maximum in the blastula,gastrula phase.The accumulation of the soluble proteins was preparing for embryonic development in the initial phase.With the development of organ formation,a large amount of the soluble proteins are consumed to provide nutrition for the embryonic metabolism.The composition of soluble proteins changed from the high molecular mass to low molecular mass during the embryonic development.The electrophoresis band of 37 kDa disappeared in heizhu phase.The electrophoresis band of 47 kDa arose from the organogenesis phase and its expression level increased significantly in the heizhu phase and inversion phase.The electrophoresis band of 44.5 kDa increased from the heizhu phase.A new electrophoresis band of 41 kDa arose in the inversion phase.The embryos hydrolyzed the high molecular proteins to the low molecular proteins of 34~52 kDa,because the soluble proteins were used to provide material and energy for the embryo development in the rap
