摘 要:从雪花莲(Galanthusnivails) 叶片中分离出了长度为484bp 的雪花莲外源凝集素cDNA,包含了完整的雪花莲外源凝集素编码序列。通过限制性酶切图谱及DNA序列分析, 从中筛选出5 种核苷酸且氨基酸序列均有差别的外源凝集素基因GNA11、GNA12、GNA13、GNA14和GNA15。将这5 个克隆与6个已报道的序列进行比较, 发现其中GNA14 与GNA15同源性最高,它们之间核苷酸序列及推导出的氨基酸序列的同源性分别为980% 和962% , LECGNA1 与GNA11 同源性最低, 它们之间核苷酸序列及推导出的氨基酸序列的同源性分别为773% 和650% 。The GNA cDNA was amplified from total snowdrop leaves RNA by RT PCR, and was cloned into EcoRV site of pBluescript KS(+). DNA sequence analysis indicates that the cloned 484 bp DNA fragment carry the entire GNA coding sequences. Five clones, GNA11, GNA12, GNA13, GNA14 and GNA15, were selected and compared with six published genes sequence. The highest homology was found between GNA14 and GNA15, with the similarities of 98 0% and 96 2% at the nucleotide level and at the deduced amino acid level, respectively. LECGNA1 and GNA11 showed the lowest homology of 77.3% and 65.0% for the nucleotide sequence and the amino acid sequence.
