摘 要:利用ISSR分子标记技术对引入我国的23个一品红(Euphorbia pulcherrima Willd.)品种基因组DNA进行扩增,分析其遗传多样性和亲缘关系。结果表明:利用筛选出的6条引物,共扩增得到100条清晰可辨条带,其中多态性条带90条,多态性位点比例为90.00%,表明一品红种质资源存在丰富的遗传多样性。6条引物在23份种质中检测到数目不等的特异条带,这些特异条带对鉴别一品红品种具有一定的价值;23个品种的相似系数为0.556 6~0.924 5,平均相似系数为0.795 0,说明一品红品种亲缘关系较近。聚类分析结果表明:国内引入的主要一品红资源的遗传关系大体上符合种苗来源差异,来自Fischer公司的"火星"与其他品种遗传距离最大,亲缘关系较远,可能具有较强的杂种优势。A polymorphism amplification of the genomic DNA of 23 poinsettia(Euphorbia pulcherrima Willd.) cultivars with ISSR molecular markers was conducted to evaluate their genetic diversity and relationship.A total of 100 clearly identifiable bands,including 904 polymorphic bands,were obtained through amplification with 6 primers.The average percentage of polymorphic locus was 90.00%,which indicated the high genetic diversity of poinsettia germplasm resources.Different quantities of specific bands were detected by 6 primers from 23 germplasms,which could be used to identify the specific poinsettia germplasm.There was high similarity between poinsettia germplasm resources,as indicated by the similarity coefficient of 23 poinsettia germplasms between 0.556 6 to 0.924 5,with an average of 0.795 0.Clustering results suggested that the genetic relationship between the major poinsettia resources generally consistented with differences in seed sources,the genetic distance between "Mars"and other cultivars was largest and the relationship was distant,so it might have strong heterosis.
