补肾活血方对成骨细胞生长因子TGF-β_1 mRNA表达的影响

摘　　要：目的：探讨补肾活血方对成骨细胞转化生长因子－ β１ （ＴＧＦ－ β１ ） ｍ ＲＮＡ表达的影响。方法：将体外分离培养的新生ＳＤ大鼠的颅骨成骨细胞，用不同浓度的骨愈片药液（１００μｇ～１００００μｇ·ｍ ｌ－ １ ）进行处理。２４小时后，利用制备的地高辛标记的鼠ＴＧＦ－ β１ ｃＤＮＡ探针进行成骨细胞的核酸原位分子杂交分析，以其细胞内杂交阳性颗粒的平均光密度值代表ＴＧＦ－ β１ ｍ ＲＮＡ的表达。结果：随着骨愈片药液浓度的增加，成骨细胞内ＴＧＦ－ β１ ｍ ＲＮＡ表达明显高于对照组。结论：补肾活血方可刺激成骨细胞ＴＧＦ－ β１ ｍ ＲＮＡ 的分泌和合成，有利于新骨生长。Objective:To investigate the effects of Prescription for Invigorate the kidney and Promote blood circulation on expression of transforming growth factor-β 1(TGF-β 1) in rat osteoblasts.Method:Primary culture and subculture of newborn SD rat's calvarial osteoblasts were estalished in vitro fist and then Guyu tablet (GYT) concentrated form 100μg to 10000μg·ml -1 was added to the medium.After 24h of treatment,in situ nucleic acid hybriodzation to rat TGF-β 1 was performed using digoxigenin-labelled TGF-β 1 cDNA probes. The mean OD value of the positive staining by hybridization measured by image anatysis represented the expression of both growth factor mRNA. Result:We found that the high level of TGF-β 1 expression in osteoblast was caused by GYT in dose dependent manner.Conclusion:GYT could stimulate the secretion and the synthesis of TGF-β 1 in osteoblast and promote the new bone proliferation.
