胆固醇缺乏对Jurkat细胞周期调控蛋白表达的影响

摘　　要：本研究从细胞周期调控这一角度来探讨胆固醇缺乏对Ｔ淋巴细胞增殖影响的分子机制。结果显示：经去脂血清培养基培养的Ｊｕｒｋａｔ细胞加ｌｏｖａｓｔａｔｉｎ 处理３ｄ 后，３Ｈ－ＴｄＲ掺入率明显下降，免疫细胞化学染色及免疫印迹法分析表明处理后的细胞ｃｙｃｌｉｎ Ｄ１、ＣＤＫ４及ｐＲｂ 蛋白的表达明显降低，加入低密度脂蛋白（ＬＤＬ）可以部分逆转上述变化。结果提示，无论是内源性还是外源性胆固醇的缺乏都能抑制Ｊｕｒｋａｔ细胞的增殖，推测这种变化可能与ｃｙｃｌｉｎ Ｄ１、ＣＤＫ４及ｐＲｂ 蛋白表达量的下降而导致相应的ｃｙｃｌｉｎIn this report, the molecular mechanism of cholesterol deficiency affecting proliferation of T lymphocytes was studied from the angle of cell cycle control. The results showed that the DNA synthesis capacity was decreased and the expression of cyclin D1,CDK4 and rRb proteins, which are the key regulators of G 1→S phase transition, was down regulated in Jurkat cells after treated with lovastatin for 3 days. When supplied with LDL, the cellular proliferation and the expression of these cell cycle regulatory proteins were partly increased. These results suggest that the down regulation of cyclin D1, CDK4 and pRb proteins may be related to the effects of cholesterol deficiency on proliferation of Jurkat cells.
