摘 要:为培育耐碱的杂种砧木,以耐碱品种资阳香橙(Citrus junos Sieb.ex Tanaka)与不耐碱的枳[Poncirus trifoliata(L.)Raf.]杂交,将杂种F1群体经形态学与SSR鉴定杂种真实性,并通过叶片黄化程度分级、叶绿素与有效铁质量分数的测定以筛选耐碱杂种单株。结果表明,(1)与母本资阳香橙叶片形态上无明显差异的F1都是珠心苗。(2)真实杂种苗从第10片叶开始都有三出复叶。(3)筛选到2对SSR引物能鉴定出全部杂种苗。(4)在碱性土上,耐碱杂种苗叶绿素cha+b的质量分数显著低于耐碱亲本资阳香橙,而极显著高于不耐碱亲本枳;但叶绿素a的质量分数显著高于父母本。5、耐碱杂种叶片中有效铁质量分数显著高于枳。研究获得了2株耐碱能力较强的真实杂种苗。In order to obtain high tolerance rootstocks to calcareous soil from citrus inter-genera offspring,hybridization were made with artificial cross-pollination method between Ziyangxiangcheng(Citrus junos Sieb.ex Tanaka) and Trifoliate orange [Poncirus trifoliata(L.) Raf.].The zygotic offspring were identified both by visible morphological characteristics and SSR markers.After growing on calcareous soil for one year,the tolerance capability of zygotic offspring was evaluated based on the degree of leaf yellowing from no yellowing symptom to extreme yellowing.The contents of chlorophyll and active iron in leaves were also analyzed.Two SSR primers were selected to identify all zygotic offspring.The result indicated that the first trifoliate leaflet usually appeared on the second leaf bud to the tenth leaf bud of the true zygotic offspring.After growing on the alkali soil of pH 8.1 for 2 years,the total contents of leaf chlorophyll a and b of the tolerance hybrids were significantly lower than Ziyangxiangcheng but significantly higher than trifoliate orange.At the same time the contents of chal a were higher than both parents.Meanwhile the contents of active iron in leaves of the tolerant hybrids were higher than trifoliate orange.Two hybrid seedlings with high tolerance to alkali soil were screened out by far.
