摘  要:目的探讨感觉神经损伤性盐敏感性高血压大鼠的氧化应激反应。方法 28只雄性Wistar大鼠分成4组:对照组、高盐组、辣椒辣素组和辣椒辣素高盐组,出生后第1、2 d 2次皮下注射辣椒辣素(CAP)50 mg/kg,动物在哺乳期结束时,高盐组给予高盐饲料(含4%NaCl),制备感觉神经损伤性盐敏感性高血压大鼠,连续4周;检测大鼠尾收缩压、血浆丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力、一氧化氮(NO)含量和一氧化氮合酶(NOS)活力。结果与对照、高盐、辣椒辣素组比较,辣椒辣素高盐组大鼠尾收缩压[(151.43±4.20)mm Hg]、MDA含量[(2.14±0.51)nmol/mL]明显升高,差异有统计学意义(P<0.05);辣椒辣素高盐组SOD活力[(185.04±3.62)U/mL]、NO含量[(49.12±3.86)μmol/L]和NOS活力[(16.99±2.98)U/mL]明显降低(P<0.05)。结论感觉神经损伤性盐敏感性高血压大鼠存在氧化应激反应。Objective To explore oxidative stress of salt-sensitive hypertension induced by sensory denervation.Methods Male Wistar rats were given 50 mg/kg capsaicin subcutaneously on the 1st and 2nd day of life.The control rats were treated with vehicle.After weanling period,the rats in treated and control groups were subdivided into 2 groups,respectively,and fed different salt diets for 4 weeks(control,high sodium diet,capsaicin,and capsaicin plus high sodium diet).The model of salt-sensitive hypertension induced by sensory denervation was established.The plasma content of malondialdehyde(MDA) and nitrogen monoxide(NO),the plasma activity of superoxide dismutase(SOD) and nitric oxide synthase(NOS) were tested.Results The tail-cuff systolic blood pressure(mmHg)of capsaicin plus high sodium group increased significantly(151.43 ±4.20).The content of MDA were significantly higher in capsaicin plus high sodium group(2.14±0.51 nmol/L) than that of the control group,capsaicin group and high sodium group(P0.05 for all).The plasma activity of SOD(185.04±3.62 U/ml)and NOS(16.99±2.98 U/ml),and the plasma content of NO(49.12±3.86 μmol/L) were all significanatly lower in capsaicin plus high sodium group than that of the control group and high sodium group(P0.01).The indexes also decreased in capsaicin plus high sodium group than in high sodium group(P0.05).Conclusion There is oxidative stress in the rats with salt-sensitive hypertension induced by sensory denervation.
