摘 要:目的:研究和厚朴酚对急性肝炎的肝保护作用。方法:复制伴刀豆球蛋白A(ConA)诱导的小鼠自身免疫性肝炎模型,并在复制模型后2h给予和厚朴酚干预。采用临床自动生化分析仪检测不同时间点丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)变化;苏木精-伊红染色法(HE)染色组织病理学观察,评价和厚朴酚治疗效果;原位末端标记法(TUNEL)染色观察8h小鼠肝脏细胞凋亡情况,定量聚合酶链式反应(PCR)检测核因子κB(NF-κB)的信使核糖核酸(mRNA)表达水平。结果:和厚朴酚可明显降低ConA诱导急性肝炎模型小鼠血清AST、ALT含量(P<0.01或P<0.05),明显减轻急性肝炎小鼠的肝组织损伤和炎性细胞浸润;ConA+和厚朴酚高、中、低剂量组可显著降低NF-κB的mRNA转录水平(P<0.05)。结论:和厚朴酚能显著减轻ConA诱导的自身免疫性肝炎的肝功能损伤,抑制NF-κBmRNA转录水平。OBJECTIVE: To investigate the protecting liver effect of honokiol in an experimental model of acute hepatitis. METHODS: Mice were injected with concanavalin A (Con A) to establish autoimmune hepatitis model. Model mice were treated with honokiol 2 h after modeling. The levels of ALT and AST were assessed by clinical automatic biochemical analysis. HE stain was used to observe histopathological change and evaluate therapeutic efficacy of honokiol. TUNEL assay was adopted to observe 8 h apoptosis of liver cells. The mRNA expression levels of NF-κB were also determined by real time PCR. RESULTS: The serum level of ALT and AST were significantly decreased by honokiol in mice with acute hepatitis induced by ConA (P〈0.05 or P〈 0.01 ). Honokiol could relieve injury of liver tissue and the infiltration of inflammatory cells in mice with acute hepatitis significantly. The mRNA expression levels of NF-κB in ConA+high-dose, medium-dose and low-dose honokiol groups decreased significantly (P〈0.05). CONCLUSION: Honokiol markedly relieves the injury of liver tissue in mice with autoimmune hepatitis induced by Con A and inhibits the mRNA expression levels of NF-κB.
