鳖甲煎丸对肺纤维化模型大鼠外周血CTGF表达的影响

摘　　要：目的:探讨鳖甲煎丸对肺纤维化模型大鼠外周血结缔组织生长因子(CTGF)表达的影响。方法:SD大鼠50只随机分为生理盐水对照组、模型对照组、醋酸泼尼松组、鳖甲煎丸中剂量组、鳖甲煎丸高剂量组,采用博莱霉素气管内注入制备肺纤维化模型,造模后各组分别用生理盐水、西药、中药灌胃,第3、14、28天乙醚麻醉后毛细玻管眼眶静脉丛采血,ELISA法检测CTGF表达明显减少,与模型对照组比较有显著性差异(P<0.05)。结果:模型对照组外周血中CTGF表达明显增加,第14天鳖甲煎丸中、高剂量组外周血CTGF的表达。结论:鳖甲煎丸可抑制肺纤维化模型大鼠外周血CTGF的表达,可能是抗肺纤维化作用的机制之一。Object： To study the effect of Biejiajian Pills on expression of CTGF（connective tissue growth factor,CTGF） in peripheral blood of the pulmonary fibrosis model rats.Method： 50 SD rats were randomly divided into 5 groups： saline control group,model control group,prednisone acetate group,middle dosage of Biejiajian Pills group and high dosage of Biejiajian Pills group.After the pulmonary fibrosis model was established by intratracheal instillation of bleomycin,each group was given normal saline,western medicine and Chinese medicine by gavage respectively.At the time point of the 3rd day,the 14th day and the 28th day,blood samples were collected from orbital venous plexus with glass capillary method for the examinaton of serum CTGF with ELISA method after the rats were anesthetized by inhalation of ether.Result： The expression of CTGF in peripheral blood of model control group increased obviously.At the time point of the 14th day,the expression of CTGF of middle dosage of Biejiajian Pills group and high dosage of Biejiajian Pills group decreased obviously.Compared with that of the model group,there was significant difference（P0.05）.Conclusion： Biejiajian Pills could inhibit the expression of CTGF in peripheral blood of the pulmonary fibrosis model rats,which probably is one of the mechanism of anti-fibrosis effect.
