摘  要:目的研究亚砷酸钠(NaAsO2)对上皮细胞向间质细胞的转变(epithelial to mesenchymal transition,EMT)过程的影响。方法①EMT模型的建立:培养小鼠乳腺上皮细胞(NMuMG),当细胞大约融合50%时,换成3%血清的培养基,24h后加入5ng/ml TGF-β,作用24 h后,观察NMuMG形态,细胞染色检测E-ca、Vimentin表达、实时定量RT-PCR检测E-ca,Vimentin mRNAs表达水平,是否成功的由上皮细胞转变成间质细胞。②加入NaAsO2:以5 ng/ml TGF-β作为阴性对照组,观察组5ng/ml TGF-β+5μmol/ml NaAsO2、5ng/ml TGF-β+10μmol/ml NaAsO2,再次检测上述指标。结果①成功建立EMT模型:细胞形态由立方形的上皮细胞转变成长梭形间质细胞;细胞染色显示,与未诱导组相比,E-ca表达呈阴性,Vim-entin表达明显呈阳性;E-ca基因表达水平显著下降,Vimentin基因表达水平显著上调。②NaAsO2对EMT过程的影响:一定浓度范围内,随NaAsO2浓度升高,明显增强了Vimentin表达;E-ca基因水平逐渐降低,Vimentin基因水平明显升高,呈剂量-效应关系。结论 NaAsO2促进了EMT过程,可能是砷中毒引发癌症的发病机制之一。Objective To explore the effect of sodium arsenite on the epithelial to mesenchymal transition(EMT) process.Methods NMuMGs were cultured,when it grew up to 50%,3% serous medium were changed,and 5 ng/ml TGF-β was added into medium after 24 hour.When 5 ng/ml TGF-β working on the NMuMGs for 24 hours,the NMuMGs had shown spindle mesenchymal cell.5 ng/ml TGF-β was as negative control,and 5 ng/ml TGF-β+5 μmol/ml NaAsO2,5 ng/ml TGF-β+10 μmol/ml NaAsO2 were observed.Results The cubical epithelial cell showed on spindal mesenchymal cell,the expression of E-ca in induced group was negative,and the expression of Vimentin was positive;the expression of E-ca mRNA in induced group was reduced,and the expression of Vimentin mRNA was increased.Meanwhile,the mRNA of E-ca was decreased and Vimentin was increased following exposure to 5 ng/ml TGF-β.When NaAsO2 were added,the cellular appearance was similar to control group,but the mRNA of E-ca and Vimentin had significant difference.Conclusions Sodium arsenite promotes EMT process which maybe the mechanism of arseniasis cancer.
