摘 要:目的:观察消水肿Ⅱ号对亚硝酸左旋精氨酸甲酯(L-NAME)7mg/kg致妊娠高血压综合征(妊高征)大鼠血压、尿蛋白及胎盘胎仔发育的影响。方法:雌雄鼠合笼次日晨,雌鼠阴道涂片发现精子确定为孕0d(E0d),按照顺序依次分入各受试组,即正常孕鼠组(A组,n=6,E10d开始灌胃蒸馏水10mL/kg)、妊高征模型对照组(B组,n=10,E10d灌胃蒸馏水10mL/kg)、硫酸镁阳性对照组(C组,n=8,E14d开始腹腔注射硫酸镁注射液100mg/kg)、消水肿Ⅱ号42g生药/kg组(D组,n=10,E10d开始灌胃消水肿Ⅱ号42g/kg)、消水肿Ⅱ号21g/kg组(E组,n=9,E10d开始灌胃消水肿Ⅱ号21g/kg)。孕期14d开始,除A组外其他各组经静脉注射L-NAME7mg/kg1次/d,共4d。测定大鼠尾动脉血压,尿蛋白含量,胎仔、胎盘质量及死胎率、畸胎率。结果:实验结束时,D组大鼠收缩压,舒张压,平均动脉压与B组比较,差异有统计学意义(均P<0.01)。E组较B组收缩压明显降低(P<0.05)。消水肿Ⅱ号对尿蛋白含量未见明显改善作用。E组死胎率较妊B组明显降低,畸胎率亦显著下降(均P<0.01)。结论:消水肿Ⅱ号能够有效降低妊高征大鼠血压,促进妊高征大鼠胎盘供血和胎仔发育。Objective: To study the hypotensive and preventive effects of XSZⅡ on pregnancy-induced hypertension in rat model established by L-NAME. Methods: Pregnancy rats were assigned to 5 groups according to the sequence of pregnancy including normal pregnancy rats (A group, n = 6, PO distilled water 10 mL/kg from E10),hypertension control rats (B group, n = 10, PO distilled water 10 mL/kg from E10),Epsom salt control rats (C group,n = 8, ip Epsom salt 100 mg/kg from E14),XSZⅡ 42 g/kg group(D group,n = 10, PO XSZⅡ 42 g/kg from 10) and XSZⅡ 21 g/kg group(E group,n = 9, PO XSZⅡ 21 g/kg from E10). Pregnancy rats were injected 7 mg/kg L-NAME i.v. on the E14 once a day for 4 days except for rats of A group. The values of tail arterial blood pressure, urine protein, placental weight, fetal death rate and birth defects rate were measured in rats. Results: There were different significances in systolic blood pressure, diastolic blood pressure and mean arterial pressure between D and B group(P 0.01). The systolic blood pressure was significantly decreased in E group compared with that of B group (P 0.05). There was no significant improvement in urine protein in rats treated by XSZⅡ. The still birth rate was significant lower in E group than that of B group (P 0.01). The abnormal embryo rate was also significant lower in rats of E group than that of control group (P 0.01). Conclusion: For pregnancy-induced hypertension, XSZⅡ can effectively reduce the blood pressure and promote blood supply to the placenta and fetal rat development.
