摘  要:目的探讨SiO2能否在体外诱导人支气管上皮细胞(HBE)发生上皮-间质转型(EMT)。方法以HBE为研究对象,0、50、100、200、300μg/mlSiO2处理HBE,倒置显微镜下观察细胞形态学改变,免疫印迹(Westenblot)法检测E-钙黏蛋白(E—cad)、α平滑肌肌动蛋白(α-SMA)及波纹蛋白(Vim)的表达,体外划痕试验检测细胞迁移能力。结果SiO2处理HBE细胞后,细胞间隙增宽,形态上呈纺锤形、梭形成纤维细胞样改变。随着SiO2浓度的升高,E—cad表达逐渐降低,300μg/ml组E-cad表达最低,为对照组的-2.98倍,100、200、300μg/ml组与对照组的差异均有统计学意义(P〈0.05)。Vim表达逐渐上调,300μg/ml组Vim表达最高,是对照组的4.46倍,200、300μg/ml组与对照组的差异有统计学意义(P〈O.05)。在0~200μg/mlSiO2浓度范围内,随着Si02浓度的升高,α—SMA表达逐渐上调,200μg/ml时α—SMA表达最强,是对照组的3.55倍,在300txg/ml时α-SMA表达略有下降,为对照组的1.90倍,100、200、300μg/ml组与对照组的差异均有统计学意义(P〈0.05)。SiO2处理组细胞体外划痕愈合面积划痕面积百分比明显大于对照组,差异有统计学意义(P〈0.05)。结论SiO2可诱导HBE细胞发生上皮-间质转型。Objective To investigate SiO2-induced EMT in human bronchial epithelial cells HBE in vitro. Methods HBE cells were cultured and then stimulated with indicated doses of SiO2 (0, 50, 100, 200, 300 μg/ml). The morphological changes were observed by microscope. In addition, Western blot was performed to detect the expression of E-cad, α-SMA and Vim. The changes of migration ability were examined by wound-healing assay in vitro. Results ( 1 )After exposure to SiO2, HBE cells lost contact with their neighbor and displayed a spindle-shape, fibroblast-like morphology. (2)Compared with the control, the E-cad (300 μg/ml group) expression downregulated 2.98 fold (P〈0.05), and the Vim (300 p.g/ml group) and α-SMA (200 μg/ml group) expression upregulated 4.46 fold and 3.55 fold (P〈0.05). There were significant differences between 100, 200, 300 μg/ml groups and the control group (P〈0.05). (3) In the test group, the percentage of woundhealing areas/wound areas were larger than those in control group (P〈0.05). Conclusions SiO2 could induce EMT in human bronchial epithelial cells.
