摘  要:MADS-box基因是真核生物中一类重要的转录调控因子,调节着植物根、茎、叶到花的发育和果实的成熟。本研究以中棉所36花发育期均一化全长cDNA文库为基础,分离出一个棉花的MADS-box基因全长cDNA命名为GhMADS12。同时利用pBI121双元载体成功构建了GhMADS12基因的植物过表达载体,并利用基因枪轰击法瞬时表达载体,验证了载体的可靠性,为下一步转基因验证基因的功能奠定了基础。MADS-box genes are an important class of transcription factors.MADS-box genes control diverse developmental processes in flowering plants ranging from root to flower and fruit development,especially control the processes of the transition from vegetative to reproductive development and establishment of floral organ identity.Based on the full-length normalized cDNA library for the flower development stages of the Chinese Cotton Research Institute 36 short-season cotton(Gossypium hirsutum L.),the full-length cDNA of a MADS-box gene,namely GhMADS12,was acquired.An over-expression vector including GhMADS12 gene and GUS gene was constructed based on pBI121,and using particle bombardment to verify the reliability of the recombinant vector.This is the basis for further gene function validation of transgenic.
